论文标题:絮凝剂在维生素C生产中的应用研究 Studies on Aplication of Flocculation in the Manufacture of Vitamin C 论文作者 张英人 论文导师 孙多先,论文学位 硕士,论文专业 制药工程 论文单位 天津大学,点击次数 88,论文页数 57页File Size564k 2003-06-01论文网 http://www.lw23.com/lunwen_126498897/ 古龙酸;絮凝;超滤 gulonic acid ; flocculate ;superfiltration . 古龙酸作为维生素C生产中的重要中间体,其质量的提高,对于维生素C的内在质量(包括晶型的完整、均一、保质期的延长等)有着至关重要的作用,本论文考察了以絮凝法对发酵液进行预处理,效果较好。采用絮凝沉淀工艺,并利用紫外吸收分光光度法测定了絮凝后渣液的重量和上清液的吸光度,确定了用壳聚糖做主凝剂,用聚丙烯酰胺做助凝剂作为发酵液预处理试剂。本论文考察了以絮凝后的上清夜进行超滤,比较超滤前后透析液和上清液蛋白的去除率,确定了最佳的操作条件和清洗条件。实验研究表明,在同一批发酵液其物化性能相同的条件下,影响絮凝效果的主要因素为絮凝剂、温度、发酵液pH值。以上清液中古龙酸的含量、渣液重量、上清液吸光度为考察指标,采用正交试验及极差分析方法,研究这些因素对上述指标的影响,而且得到了最佳絮凝条件:温度40℃、絮凝剂加入浓度2000ppm、pH7.0、时间4h;同时研究了影响超滤操作和清洗效果的因素是操作压力、温度、操作方式。采用正交试验及极差分析方法研究这些因素对上述指标的影响,而且得到了最佳操作条件:压力50—70KPa,温度40℃以下,压差100 KPa。同时发现采用滤液脉冲反洗操作(反冲压力150 KPa,反冲时间2—4秒,反冲周期5分钟),在此分离条件下,古龙酸的收率为88.5%,比原工艺提高收率9%。 Improving the quality of the gulonic acid which is a intermediate of vitamin c , is important to the inter-quality of vitamin c.(including crystal integrity ,order and quality extension etc.)In the thesis flocculent method as preparing treatment has better effect. Adopting deposition method and ultra-violet absorption spectrometry measure the weight of dreg and the absorbency of upper pure liquid, recognize the chitosan (main flocculent reagent) and polyacrylonitrile amide (vice flocculent reagent) as preparing treatment reagent for the ferment liquid.In the thesis superfiltrating the upper pure liquid after flocculent ,comparing the rate of wiping of the dialysis liquid before and back the superfiltration and the albumen of the upper pure liquid, conclude the optimum condition of the operating and the purgation . The experiment research expresses that under the same physical and chemical condition in same batch ferment liquid ,the main factors affect the flocculent are flocculent reagent ,temperature , the pH value of the ferment liquid. Being the index of the content of the gulonic acid , the weight of dreggy liquid , and the absorbency of upper pure liquid, adopting the orthogonal method and the error analysis method , studying the factors affecting these indexes , conclude not only the optimum condition of the flocculent:temperature 40℃,concentration 2000ppm,pH7.0, time 4h; and the factors affecting superfiltration and purgation are operating pressure ,temperature, working way ; but also the optimum working conditions: pressure 50-70kpa , temperature 40℃ below, differential pressure 100kpa.And at the same time adopting filtrate pulse anti- wash working (anti-buck compressive force 150kpa ,anti- bulk time 2-4seconds , anti-bulk period 5minutes), the collect rate of gulonic acid is 88.5% , 9 percent higher than original technics .
