论文标题:血小板源性生长因子与妊娠期高血压疾病的相关性研究 Research on the Relationship between Platelet-derived Growth Factors and Hypertensive Disorder Complication Pregnancy 论文作者 论文导师 杨茵,论文学位 硕士,论文专业 妇产科学 论文单位 福建医科大学,点击次数 100,论文页数 45页File Size1282K 2007-03-01论文网 http://www.lw23.com/lunwen_138499932/ Platelet-derived growth factor ;; Hypertensive disorder complication pregnancy;; Artherosclerosis;; Placenta 目的:通过比较正常孕妇和妊娠期高血压疾病患者胎盘组织中PDGF-A、-B、-C和-D的表达以及血清PDGF-AA和-BB水平,探讨PDGFs与妊娠期高血压疾病发病的关系。 方法:入选2005年8月-2006年6月入院的轻、重度子痫前期患者各20例,子痫患者3例,另随机选择年龄段、孕周相应的正常孕妇20例作为对照;采用逆转录聚合酶链反应(RT—PCR)检测3组妊娠期高血压疾病患者和20例正常妊娠孕妇胎盘绒毛组织PDGF-A、-B、-C和-DmRNA的表达;分别采用免疫组织化学方法和酶联免疫吸附试验(ELISA法)分析各组胎盘与血清中PDGF-AA和PDGF-BB的蛋白表达量。 结果:(1)RT-PCR显示:妊娠期高血压疾病及正常妊娠胎盘绒毛组织均有PDGF-A、PDGF-B、PDGF-C和PDGF-DmRNA表达,但妊娠期高血压疾病患者胎盘绒毛组织PDGFs表达均明显高于正常对照组,且与病情轻重呈正相关(P均<0.01,;(2)免疫组化提示:妊娠期高血压疾病患者盘绒毛组织PDGF-AA和PDGF-BB蛋白的表达均明显高于正常对照组,且与病情轻重呈正相关(P均<0.01);(3)ELISA法示妊娠期高血压疾病患者血清PDGF-AA和PDGF-BB蛋白水平均明显高于正常对照组,且与病情轻重呈正相关(P均<0.01)。 结论:(1)PDGF-A、PDGF-B、PDGF-C和PDGF-D在正常孕妇及妊娠期高血压疾病患者的胎盘均有表达,表明PDGFs可能是正常妊娠所不可缺少的重要因子;(2)妊娠期高血压疾病患者PDGFs的表达均明显高于正常对照组,且与病情轻重呈正相关,表明PDGFs与妊娠期高血压疾病的发生和发展密切相关,高度表达的PDGFs可能通过促进动脉粥样硬化在妊娠期高血压疾病的发病中起着重要的作用。 Objective: To compare the expression of platelet-derived growth factor (PDGF) -A, -B, -C and -D in the placentas and serum PDGF-AA and -BB from patients with hypertensive disorder complication pregnancy with those from normal pregnant women so as to explore the relationship between PDGFs and the pathogenesy of hypertensive disorder complication pregnancy. Methods: From August 2005 to June 2006, 43 patients with hypertensive disorder complication pregnancy were selected in our study. Another 20 age- and gestational week-matched normal pregnant women served as controls. Reverse transcription -polymerase chain reaction (RT-PCR) were applied to determine the mRNA expression levels of PDGF-A, -B, -C and -D in the placentas from the 4 groups; Meanwhile, the serum levels of PDGF-AA and -BB as well as their expression in the placentas from the 4 groups were determined with immunohistochemistry and enzyme linked immunosorbent assay (ELISA), respectively. Results: 1)RT-PCR showed that all subjects expressed PDGF-A, -B, -C and -D in the placentas, while the mRNA expression levels of the four kinds of PDGFs in the patients with hypertensive disorder complication pregnancy were significantly higher than those in the normal pregnancy group(P all<0.01), and were positively correlated with the degree of the disease (2) Compared with the normal pregnancy group, significantly higher expression levels of PDGF-AA and -BB were observed in the placentas of patients with hypertensive disorder complication pregnancy by immunohistochemistry (P all<0.01), and were positively correlated with the degree of the disease. (3) ELISA showed the serum levels of PDGF-AA and -BB in the patients with hypertensive disorder complication pregnancy were significantly higher than those in the normal pregnancy group(P all<0.01), and were positively correlated with the degree of the disease. Conclusions: (1) PDGF-A, -B, -C and–D express in the placentas of both patients with hypertensive disorder complication pregnancy and normal pregnant women, which shows that PDGFs may be necessary cytokines for normal pregnancy. (2) Patients with hypertensive disorder complication pregnancy have higher expression levels of PDGF-A, -B, -C and–D than normal pregnant women, with the expression levels positively correlated with the degree of the disease, which shows PDGFs play an important role in the development and progression of hypertensive disorder complication pregnancy by promoting atherogenesis in the placental vessels.
