本论文结合春石斛在北京地区的盆花生产,对春石斛的扦插繁殖技术、有性繁殖技术中的无菌播种、以及花期调控中花芽分化等技术进行了初步研究,得到了以下结论: 1.扦插繁殖技术试验结果表明: ①D.Tomoflake‘Akebono’:不适宜使用NAA,对照可使出芽率高达73%,株高9.010cm,生根数4.25条,根长2.3607cm,根粗0.1387cm,但是成活率在NAA150mg/L处理时最高,达83.3%。D.Shigiriya lady:较为适宜的浓度为NAA100mg/L,出芽率达81%,株高7.980cm,生根数5.15条,根长1.4726cm,根粗0.2026cm,在50mg/L处理时成活率最高,达70%。 ②D.Tomoflake‘Akebono’:6-BA 200mg/L处理最优,出芽率为52.1%。D.Shigiriya lady:6-BA 150mg/L处理最优,出芽率高达79.2%。 ③6-BA200mg/L浸泡处理对春石斛D.Tomoflake‘Akebono’假鳞茎平铺最优,出芽率最高达55.0%。 ④D.Shigiriya lady茎段扦插时选用中下部的茎段为宜。出芽率达76.7%以上。春夏季扦插的出芽时间要比秋季扦插提前30天。春夏季扦插时,出芽率只有51.6%,而秋季扦插时可达93%。 2.人工杂交试验结果表明: 经过人工授粉, 6-8个月果实逐渐成熟,结实率平均为62.1%,D.Tomoflake‘Akebono’自交可以结实,结实率为36.4%。选择较为健壮的植株作为母本,则正交的结实率高于反交的结实率。果荚长度介于2.6cm到3.77cm之间,宽度介于1.00cm到1.54cm之间。每一滴无菌水中所含的种子数大约从20到100粒不等,有胚数从0.2到9.4不等,有胚率高可达77.81%,低的不足10%。不同的杂交组合结实情况与父母本的性状、成熟度等都有关系, 3.无菌播种试验结果表明: 春石斛无菌播种的试验是最适宜的培养基:改良的N6培养基+6-BA0.2mg/L+NAA0.5mg/L+椰汁50mg/L。固体培养基条件下, 4周方可肉眼观察到原球茎的膨大, 6周后可观察到萌发,液体培养3周后即可观察到原球茎的膨大。相对于静置培养来说,振荡培养原球茎萌发得早,增大的快。适宜种胚苗生长的培养
The key of cutting, aseptic seeding,regulation of florescence of pot Nobile-type Dendrobium production were studied in this paper.The results can be summaried as follows: 1. Cutting: For D.Tomoflake‘Akebono’:the suitable concentration of NAA was 0mg/L.The bud rate with the treatment of NAA(0mg/L)was 73%,and the height of the young plant was 9.010cm,and the number of the root is 4.25,and the length is 2.3607cm,and the width is 0.1387cm.For D.Shigiriya lady:the suitable concentration of NAA was 100mg/L. The bud rate with the treatment of NAA(100mg/L)was 81%,and the height was 7.980cm,and the number of the root is 5.15,and the length is 1.4726cm,and the width is 0.2026cm. For D.Tomoflake‘Akebono’:the suitable concentration of 6-BA was 200mg/L,The bud rate is 52.1%.For D.shigiriya lady:the suitable concentration was 150mg/L.The bud rate is 79.2%. The experiment of different part of stem of D.Shigiriya lady showed that the better parts was the middle and the below ones.The bud rate of the middle one was 76.7%,and the later was 80%. Without hormone , cutting in spring and summer was 30d earlier than in autumn.While the bud rate in spring and autumn was 51.6% less than in autumn with 93%. 2. The experiment of traditional hybridization showed as follows: Whether the hybridization was succeed could be judged primarily by seeing the fruit maintained green or not. Then after one month, judging by the bulgy germen ultimately. The fruit become ripe within 6-8 months.And the fruit could be crazed after 8 months.The average rate of fructification was 62.1%. D.Tomoflake‘Akebono’could
