论文标题:电针对失眠模型大鼠睡眠免疫因子影响的实验研究 The Experimental Study on the Effect of Electro-acupuncture to the Points Immunocytokin of Insoninia Rats 论文作者 论文导师 傅杰英,论文学位 硕士,论文专业 针灸推拿学 论文单位 广州中医药大学,点击次数 115,论文页数 45页File Size2687K 2007-04-01论文网 http://www.lw23.com/lunwen_190731817/ insomnia; rat model of insomnia; electro—acupuncture; points immunocytokin 失眠是以难以入睡和睡眠维持困难为特征的一种最常见的睡眠障碍,是一种临床的常见症状,发生率非常高。目前其发病机制尚未完全明确,化学药物治疗效果欠佳。而中医针灸治疗失眠已有数千年的经验,近年来的文献报道提示,针灸对失眠症的疗效较满意。但是目前针对治疗失眠症的研究尚待完善,围绕失眠症的研究大多集中于临床研究,实验室研究特别是失眠症体液调节机制的研究相对较少。 为了探讨针刺治疗失眠症的作用机制,本课题共用Wistar大鼠40只,采取PCPA腹腔注射法复制失眠大鼠模型,用电针申脉、照海、神门、大椎、百会穴治疗方法,通过测定针刺对失眠大鼠脑内和血内免疫细胞因子IL-1、IL-6、TNF等含量的影响,探讨针刺调节失眠大鼠睡眠情况的机制。结果:1)对照组与模型组的失眠大鼠比较,两组脑内和血清中的IL-1β、IL-6和TNF-α含量无显著性差异(P>0.05)。2)西药(安定)治疗后PCPA所致失眠大鼠脑内和血清的IL-1β、IL-6和TNF-α,在原有基础上明显升高,与对照组比较差异显著(P<0.05)。3)针刺(电针)治疗后PCPA所致失眠大鼠脑内和血清的IL-1β、IL-6和TNF-α,在原有基础上明显升高,与对照组比较差异非常显著(P<0.01)。4)治疗后PCPA所致失眠大鼠脑内和血清的IL-1β、IL-6和TNF-α,在原有基础上均有明显升高,但电针组与西药组比较差异显著(P<0.05)。 结论:1)腹腔注射PCPA可使正常大鼠的睡眠-觉醒周期消失,是复制失眠大鼠模型的较稳定方法。2)电针治疗可以使PCPA所致失眠大鼠免疫细胞因子IL-1、IL-2和TNF的含量升高,与正常组比较差异非常显著(P<0.01)。3)电针和西药的治疗,均能使失眠大鼠免疫细胞因子IL-1、IL-2和TNF的含量升高,但针刺治疗作用优于安定。4)针灸调节失眠大鼠的细胞因子的紊乱,增加免疫细胞因子的含量,是穴位电针治疗失眠症获效的机制之一,这将为临床针刺治疗失眠症提供更加客观的理论依据。 Insomnia is the most common sleeping disorder characterizedby difficulty in falling asleep and sleep maintenance, aclinical common symptom which occurrence rate is very high.At present, the pathogenic mechanism of insomnia hasn"t beendefined completely, and the effect of the chemotherapy isn"tgood. But experience on the treatment of insomnia by acupunctureand moxibustion of Traditional Chinese Medicine has alreadybeen used for several thousands years, the reports in recentyears showed that the curative effect on insomnia of acupunctureand moxibustion was quite satisfactory. However, the studyaiming at treatment of insomnia needs improvement, the researcharound insomnia is mostly concentrated on the clinical research,the laboratory studies especially the studies on the AdjustmentMechanism of Human Body Fluid of insomnia are comparativelyless. In order to explore the functional mechanism of insomniatreated by acupuncture, the experiment was prepared from 40Wistar rats, establishment of rat model of insomnia was adoptedby intraperitoneal injection with PCPA(para-chlorophenylalanine), using electric needle on the pointof Shenmen (HT7), Zhaohai (EI6), Dazhui (DU14) and Baihui(DU20), inquired into the mechanism of acupunctural adjustmenton sleep status of insomnia rats by evaluating the influenceof acupuncture to the contents of cerebral and serumimmunocytokine IL-1、IL-6 and TNF etc of the insomnia rats.Results:1) The result didn"t show significantdifference(P>0.05) by comparing the contents of the cerebraland serum IL-1, IL-6 and TNF-αbetween the control group andmodel group. 2) After administration of the western medicine(DIAZEPAM), the contents of cerebral and serum IL-1,IL-6 and TNF-αof insomnia rats due to PCPA were increasedsignificantly from the original level, and showed significantdifference (P<0.05) comparing with control groups. 3) afterthe pinprick"s(electric needle) treatment, the contents ofcerebral and serum IL-1, IL-6 and TNF-αof insomnia rats duedto PCPA were increased significantly from the original level,and showed very significant difference (P<0.01) comparing withcontrol groups. 4) after the treatment, the amounts of cerebraland serum IL-1, IL-6 and TNF-αof insomnia rats dued to PCPAwere all increased significantly from the original level, butthe difference between electric needle group and westernmedicine group was significant (P<0.05). Conclusion:1) Intraperitoneal injection with PCPA can makesleep-wakefulness cycle of normal rats disappear, and which isa quite stable establishing method of rat model of insomnia. 2)Electro-Acupuncture can increase the contents of IL-1, IL-6and TNF-αof insomnia rats in cerebrum and serum dued to PCPA,and the difference was very significant comparing with thecontrol group (P<0.01). 3) the treatment by electric needleand western medicine, both can increase the concentrations ofcerebral and serum IL-1, IL-6 and TNF-αof insomnia rats duedto PCPA, but the effect of acupuncture was better than DIAZEPAM.4) Acupunctural adjustment on the cytokine disorders ofinsomnia rats or increasing the content of the immunocytokineis one of the mechanisms of effective treatment on insomnia byelectro—acupuncture, which will provide more objectivetheoretical foundation for the clinical treatment on insomniawith acupuncture.
