本课题分别采用脉冲电针和音乐声波电针,对急性躯体痛模型Wistar大鼠进行研究。选取双侧“足三里”和“三阴交”穴,以甩尾潜伏期及中枢痛相关神经元放电活动作为测痛指标,从行为学和神经电生理学的角度对比观察了两类不同电针在连续多次电针过程中镇痛效果的差异;并进一步采用免疫组织化学和原位杂交方法同步观察了大鼠脑内八肽胆囊收缩素样免疫活性物质(CCK-8-ir)和CCKmRNA的变化情况,以探讨抗阿片物质与镇痛效果之间的关系,及不同电针在对抗机体耐受方面的可能的作用机制,试图揭示音乐声波电针在克服机体产生耐受方面的相关机理。 实验结果表明:1、在行为学方面,两类电针均有镇痛作用,但音乐声波电针组的镇痛效果优于脉冲电针组;且后者的镇痛作用随电针次数增多而下降并产生电针耐受,而音乐声波电针组的镇痛作用持久、稳定,且不产生耐受。2、在神经电生理学方面,通过同时对大鼠中枢神经系统内尾核和海马中痛兴奋神经元(PEN)和痛抑制神经元(PIN)放电活动的观察所得到的数据,进一步从细胞水平上更客观地支持了行为水平上的结果。3、通过免疫组织化学方法观察到脉冲电针组大鼠脑内尾核和海马组织中CCK-8-ir阳性表达随电针次数增加而增强,而音乐声波电针组则随电针次数增加而减弱,提示CCK-8有对抗机体镇痛的作用,连续多次音乐声波电针不使脑内CCK-8生成增加,可能是其不产生机体耐受的原因之一。4、原位杂交实验显示脉冲电针组大鼠脑内CCKmRNA阳性表达增多从第二次电针后开始,而音乐声波电针组CCK的基因表达从第二次电针后开始减弱,从分子生物学角度进一步证实了CCK在电针耐受形成过程中的重要作用。 从以上的实验结果得出结论,以音乐声波作为电针的输出波形可明显克服机体产生耐受,从而提高电针的疗效,其抗耐受的机理推测可能与机体内抗阿片物质的释放减少有关,即CCK-8是参与电针耐受的物质以及影响电针疗效的原因之一。
In this research, through animal experiments on Wistar of acute somatalgia on points of Tsusanli and Sanyinjiao adopting Tail-Flick Latency and the electric-discharge of PIN and PEN as indexes, the difference of sedation with pulse-electroacupuncture in contrast to music-electroacupuncture electroaupuncturing by many times was observed from angles of behavior science and neuroelectrophysiology. And the changes of CCK-8-ir and CCKmRNA in brains of rats were further studied synchronously by means of immunohistochemistry and in-situ-hybridization in order to probe into the relations between AOS and sedation, and attempt to bring to light the probable antitolerance mechanisms of difference electroacupunctures.Result : 1. On behavior science, both of these two electroacupunctures were verified to ease pain effectively, but the music-electroacupuncture was better than thepulse-electroacupuncture. And the effect of the latter declined with the the times of electroacupuncture increasing, and even the antitolerance occurred, while which of the former was lasting, stable, and didn"t produce antitolerance. 2. On neuroelectrophysiology, it was further verified objectively though observing the electric-discharge of PIN and PEN at CPU and HPC of rats. 3. On immunohistochemistry, the result showed that the positive CCK-8-ir at CPU and HPC of rats raised with the times of pulse-electroacupuncture adding, while which reduced with the music-electroacupuncture, which showed that CCK has the founction of anti-sedation, and it is maybe one of the reasons of the mechanism of antitolerance that CCK in rats" brains didn"t increase after music-electroacupuncture by many times continuously. 4. On in-situ-hybridization, the pulse-electroacupuncture enhanced theCCKmRNA in rats" brains from the second times of electroacupuncture, while the music-electroacupuncture decreased, which further verified the important function of CCK in electroacupuncture antitolerance from angle of molecular biology.The conclusion is that the sound wave as the output-wave of electroacupuncture can overcome antitolerance notably, thus improving the electroacupuncture"s curative effect. And it was inferred that the possible antitolerance mechanisms of music-electroacupuncture concern the reduce of releasing AOS, namely, CCK-8 participates antitolerance and is maybe the reason reducing curative effect when electroacupuncture.
