论文标题:高山红景天组织培养的研究 Study on Rhodiola Sachalinensis A.Bor by Callus Culture 论文作者 南桂仙 论文导师 许明子,论文学位 硕士,论文专业 作物遗传育种 论文单位 延边大学,点击次数 262,论文页数 38页File Size2766k 2004-06-01论文网 http://www.lw23.com/lunwen_3340952/ 高山红景天;愈伤组织;激素;光温;红景天甙 Rhodiola sachalinensis A.Bor;Callus;Hormone;Photo and Temprature;Salidraside 本文以和龙市高山红景天栽培基地采集的高山红景天种子、试管苗根、茎、叶和移栽到试验田的雌、雄植株叶片为外植体,研究了愈伤组织的诱导、分化和再生苗发根适合的培养基以及愈伤组织生长的培养条件并对不同条件下培养的愈伤组织进行红景天甙含量的比较,结果表明: 1、MS基本培养基附加浓度为2.0,8.0mg/l的NAA与不同浓度的6-BA的激素组合,可得到较高的愈伤组织诱导率;浓度为1.5mg/l的2,4-D的愈伤组织诱导率高于0.5mg/l的2,4-D,特别是与浓度为5.0,8.0mg/l的6-BA配合效果更佳。 2、MS基本培养基中附加3.0mg/l的6-BA和0.2mg/l的NAA条件下得到较高分化率:1/2MS基本培养基附加1.0mg/l的IAA条件下适合再生苗发根。 3、愈伤组织生长和红景天甙积累所需的光温条件基本一致,在24℃的温度和12—14h光照长度下有利于愈伤组织生长和红景天甙的积累。 4、蔗糖对红景天甙含量的合成有影响,加3.5%蔗糖的MS培养基中生长的愈伤组织有利于红景天甙的合成。 5、由雌、雄植株叶片诱导的愈伤组织红景天甙含量有极显著差异,由雄株叶片诱导的愈伤组织红景天甙含量高于雌株。 In this paper,we studied the fittest medium of induction and differentiation on calli and culture condition of callus growth with seeds from tamed Rhodiola sachalinensis A.Bor,root,stem and leaf of tube plants and leaf of femaleand male plants in experimental field,and compared salidraside accumulation with each other.The results of experiments wereas follows:1 Good callus growth was obtained on a medium containing both NAA 2.0mg/L or 8.0mg/L,2,4-D 1.5mg/L and 6-BA 5.0mg/L or 8.0mg/L.2 Good differentiate rate was obtained on MS base medium containing both 6-BA 3.0mg/L and NAA 0.2mg/L,and good rootage rate was obtained on 1/2MS base medium containing IAA 1 .0mg/L.3, Good callus growth and salidroside accumulation were cultured under 24 and12-14h.4, Sucrose had significant effects on the accumulation of salidroside and MS medium containing 3.5% sucrose was the best.5, Salidroside accumulation of calli inducted from leaves of female and male had more significant difference,and male was more than female.
