论文标题:基于栅格的流域地理信息系统研究与开发 RAPD analysis of genetic structure in Eucalyptus.grandis populations 论文作者 何芬芳 论文导师 吴东亮;唐小明,论文学位 硕士,论文专业 生态学 论文单位 中南林学院,点击次数 101,论文页数 61页File Size4637k 2001-05-01论文网 http://www.lw23.com/lunwen_48174757/ 栅格;流域;数字高程模型;地理信息系统 Eucalyptus grandis;population;RAPD; genetic variality;genetic structure;genetic diversity 本文采用地理信息系统的方法,建立了吉县蔡家川流域的数字高程模型(DEM)。研究了该流域地表水流方向、汇流能力、子集水区的划分等水文特征,并以DEM为基础,计算了通用土壤流失方程(USLE)中的地形因子LS值,有利于该流域土壤侵蚀量的计算,把栅格化的多边形地块与DEM进行叠加,使空间数据与属性数据一起输出到通用数据库中。结果表明:在上述基础上易于对该流域进行全面而较精确的分析,并有利于进行合理的土地利用规划,对该流域的农林业生产具有重要的指导意义。 Leaf samples collected from 11 Eucalyptus grandis provenances population introduced from the geographic range of Eucalyptus grandis in the North of QLD in Australia are analysed for RAPD variation. The genomic DNA is extracted from 80 individuals leaves with 20 familes using modified CTAB. The analysis of the genomic DNA which amplified by 20 arbitrary 10-base primers obtained from 186 10-base primers reveals 201 amplified loci with 89 polymorphic loci and the proporation of polymorphic loci is 44.28%. Genetic variablity indexes are determined using 201 amplified loci including proporation of polymorphic loci (P), diversity coefflcient( DC), proporation of diversity coefficient( PDC), Nei抯 unbiased genetic identity (I) and Nei抯 unbiased estimates of genetic distance (D) .Genetic analysis of 80 individuals from 11 populations of E.grandis shows there is highly variation in genomic DNA with P values at 44.3%. The provenance of 18591 was the most one with 39.5% at P values, so is 18594 with 3 6.7%. The variability is high in population with an average of 0.6427 DC values, 7.27% PDC values and with the average of 0.1664 unbiased estimates of genetic distance. The dendrogram using two different methods shows that a high similarity is existed among populations of 18180, 18591, 18595, 17562, 18590, 18592and18594, butthereisa lower similarity between 17857 and the other populations. The study provides a scientific foundation on early selection in grandis populations.
