本文以T-DNA插入pDs1300质粒转化水稻品种中花11获得的矮化突变株系丛矮648为材料,并以中花11为对照,对其植株形态、生理生化特性及外源生长物质调控进行了研究。对丛矮648幼苗期进行GA_3、IAA、Z处理;分蘖盛期进行了GA_3的调控;分析了丛矮648的生物学特性,测定了内源激素、可溶性糖、可溶性蛋白的含量以及过氧化物酶和吲哚乙酸氧化酶活力,主要结果如下: 1丛矮648在幼苗期株高差异不显著,在分蘖盛期株高差异显著,幼苗期内源GA_(1+4)、IAA含量高于中花11,分蘖盛期内源GA_(1+4)、IAA含量低于中花11。 2丛矮648幼苗期用三种外源生长物质处理后,二叶期丛矮648对三种物质均表现出敏感性,其中以GA_3处理敏感性最大,株高和第二叶鞘增长率分别为50.1%、65.7%,其内源激素含量相应增大。不同时期和不同GA_3剂量处理试验表明,丛矮648是全生育期GA敏感型突变体。 3丛矮648分蘖盛期叶可溶性糖、可溶性蛋白差异不显著,丛矮648光合生理指标、POD、IOD酶活力差异显著。其中在分蘖盛期14d丛矮648叶、茎的POD酶活力比中花11高43.0%、16.8%,丛矮648叶IOD酶活力比中花11高77.4%。 4分蘖盛期丛矮648株高、可溶性蛋白随着GA_3处理浓度的增加而升高,丛矮648叶POD、IOD酶活力随着GA_3浓度的增加而降低,但丛矮648茎POD酶活力随着GA_3浓度的增加而增加。 上述研究结果表明,丛矮648突变株系与中花11间形态差异与植物激素、可溶性蛋白、POD和IOD酶活力有关。
Taking rice(Oryzn Satire L.) variety zhonghua11(ZH11) as control, the plantmorphological, physiological and biochemical characteristics and the regulation ofexogenous growth substances of CA648, a rice dwarf mutant line obtained bytransformation with the plasmid pDs1300 T-DNA into rice variety ZH11, werestudied in the paper. The plants of CA648 were treated with GA_3, IAA, Z and GA_3 atseedlings stage and GA_3 at mid tillering stage, then the biological characteristics wereanalyzed, and endogenous phytohormones, soluble sugar, soluble protein, theactivities of peroxidase and IAA oxidase were also determined. The main results areas follows 1. There is no significant differences in the stem length during the seedling stage butthere are significant differences during tillering stage between CA648 and ZH11. Thecontent of endogenous GA_(1+4) and IAA in CA648 is lower at the seedling stage buthigher at mid tillering stage than that in ZH11. 2. After treated with three different plant growth regulators during the seedling period,CA648 plants at 2ed leaf stage was sensitive to the three plant growth regulators, andshowed the most sensitivity towards GA_3. The stem length and the 2ed sheathelongation rate of CA648 were 50.1% and 65.7 % respectively and the correspondingendogenous phytohormonal contents were increased accordingly. The results ofdifferent dosages of GA_3 treatment at different stages indicate that CA648 is a lifeperiod sensitive mutant within the whole life cycle. 3. No significant differences of soluble sugar and soluble protein but significantdifferences of photosynthetic indexes, POD activity and IOD activity were observedat the mid tillering stage between CA648 and ZH11. 14d after mid tillering stage,POD activities of CA648 in leaf and stem was 43.0% and 16.8 % higher than that ofZH11. The IOD activities of CA648 in leaves was 77.4 % higher than that of ZH11. 4. The plant height and soluble protein in CA648 raised with the concentration of GA_3The POD, IOD activities in leaves of CA648 reduced with the concentration of GA_3,but the POD activity in the stem of CA648 enhanced with the concentration of GA_3. The above results indicate that the morphological differences between CA648 andZH11 are correlated with phytohormones, soluble protein, POD and IOD activities.
