马尾松(Pinus massoniana Lamb.)属于松科(Pinaceae)松属(Pinus L.),是我国东南湿润亚热带地区分布最广、资源储量最丰富的森林类型,也是我国南方特有的重要工业原料林树种。因此,选择马尾松树种作为遗传资源优势深度发掘与高遗传品质林木良种选育为研究对象,完全符合当前和未来林业生产发展需要与林业科技经济发展的要求。 马尾松分布区内具有众多的高山大川,地理环境条件变化多端,因长期自然选择和生殖隔离等原因,存在着极为丰富的遗传变异,这为马尾松的材性遗传改良提供了丰富的物质基础。 本研究的主要目的是利用马尾松嫩叶为材料,检测马尾松湖南林朵8号和广西111号杂交后代的均一性,采用检验DNA序列多态性的新方法,进行马尾松杂种F1代RAPD扩增谱带分析,并进行RAPD分子标记,鉴定生产应用中优良杂种F1代;为开展马尾松基因工程定向育种奠定可靠基础,以便进一步研究和探索马尾松杂种表现的DNA早期鉴别技术,缩短育种周期,为生产提供高世代优良种质,为强优势杂种大面积生产提供可靠的技术保障。 利用改良的CTAB法抽提51个F1代马尾松叶片基因组,得到的DNA杂质含量少,通过RAPD实验扩增的指纹图谱可以看出,其得到的DNA完全符合RAPD实验的要求。 从99条随机引物中筛选出具多态性的5条引物,利用这5条引物对来自湖南省桂阳马尾松实生种子园的23个F1子代进行了RAPD扩增反应,共产生35条带,其中27条带具有变异性,占扩增产物总数的77.1%可完全把种子园的23个F1子代全部鉴别出来。利用SPSS 11.0对数据进行了处理,把23个F1代的进行了聚类。
