本文采用整体动物实验和细胞实验考察月季花水提取物抗糖尿病作用及其作用机制。在整体动物水平实验中,我们采用了四氧嘧啶造成的糖尿病小鼠模型、链脲左菌素造成的糖尿病大鼠模型以及自发性KK2型糖尿病小鼠三种方法观察月季花水提物的降血糖作用,实验结果表明:月季花水提物对四氧嘧啶造成的糖尿病小鼠和链脲左菌素造成的糖尿病大鼠以及自发性KK型糖尿病小鼠的血糖均有显著的降低。在四氧嘧啶实验中结果显示月季花在8~10g生药/kg剂量时就对高血糖动物的血糖值具有明显的改善作用,在链脲左菌素造成的糖尿病大鼠实验中,月季花水提物能同时降低血糖和糖化血清蛋白水平并提高胰岛素的分泌。在细胞水平实验中,我们对大鼠胰岛细胞进行了原代培养,同时采用三种实验方法(分别是外源性一氧化氮造成的胰岛细胞损伤模型、四氧嘧啶损伤模型和胰淀素损伤模型)观察月季花水提物对胰岛细胞的保护作用,实验结果表明:月季花水提物能够改善由外源性一氧化氮造成的胰岛细胞损伤、四氧嘧啶对胰岛细胞的损伤和胰淀素对胰岛细胞的损伤,表明月季花水提物能够保护糖尿病动物受损的胰岛细胞。另外我们还采用胰岛素抵抗细胞模型观察了月季花对胰岛素抵抗的改善作用,在胰岛素抵抗的HepG2肝癌细胞实验中,月季花水提物能够改善由高胰岛素造成的胰岛素抵抗。以上实验结果证实:月季花具有明显的降血糖作用,对动物的胰岛细胞具有明显的保护作用,同时能够改善由高胰岛素造成的胰岛素抵抗。
To investigate the antidiabetic effects and the mechnism of YJH in the levels of animals and cells . we established diabetic model rats induced by streptotocin (STZ) and alloxan , while use the spontaneous diabetic KK mice , in order to investigate effect of the decreasing glucose and GSP and increasing the concentration of insulin .The results sugest YJH can significantly decrease the level of glucose and GSP while inrease the concentration of insulin in the experiments of STZ-induced rats model , In the experiments of AXN-induced rats model , YJH can significantly decrease the level of glucose, while YJH can decrease the level of glucose in the spontaneous KK mice . The studies of islet cells experiments showed that SPN , alloxan and amylin was able to significantly decrease SOD level,increase nitric oxide(NO) and MDA production , reduce insulin release at glucose stimulation , decrease DNA concentration in islet cells . When incubated with different concentrations YJH , the injury experiments of SPN-induced , alloxan-induced and amylin-induced B-cells was reversed by reducing NO and MDA production and enhancing SOD level. So it suggest that YJH may play a protective role in B-cell injury induced by SPN , alloxan and amylin-induced a significant decrease in SOD activity , inhabit insulin and DNA concentration synthesized , while increase MDA profuction and NO production . When incubated with different concentrations YJH , the injury of SPN , alloxan amylin-induced B-cells was reversed by reducing NO and MDA production and enhancing SOD level., insulin and DNA concentration . Pancreatic islet cells from SD rats were cultured in vitro. Pancreatic islet cells were incubated with exogenous nitric oxide , alloxan and amylin and different concentrations YJH . The experimental studies of HepG2 cell explored that YJH can significantly improve the IR situation induced by high content insulin .From the results of the animals , it is indicated that YJH possesses obvious protective effect against AXN and STZ induced artificial diabetic animal , at the same time YJH
