论文标题:双色茉莉组织培养技术体系研究 The Study on Technique System of Tissue Culture in Brunfesia Latifolia 论文作者 魏海龙 论文导师 石大兴,论文学位 硕士,论文专业 森林培育学 论文单位 四川农业大学,点击次数 93,论文页数 36页File Size1820k 2002-05-01论文网 http://www.lw23.com/lunwen_96243327/ 双色茉莉;组织培养;技术体系 Brunfelsia latifolia;Tissue culture;Technique system 本研究以双色茉莉(Brunfelsia latifolia)的茎段进行组织培养体系的建立,通过全面组合设计、正交设计,结合方差分析、极差分析、多重比较,着重研究双色茉莉茎段培养的最佳培养基配方及培养程序,以建立双色茉莉组培快繁体系。结果表明: 1 双色茉莉组培快繁的外植体以4月和10月的茎段好,茎段中部诱导的不定芽粗,叶片大,长势好,分化率高。 2 初代培养:培养基1/2MS+IBA0.1mg/L+NAA0.01mg/L+BA3mg/L+KT2mg/L的效果最好。 3 继代培养:培养基为MS+IBA0.5mg/L+BA3mg/L+活性炭0.2%,35天时平均增值系数为5.92,大于2cm芽率为47.16%,活性炭对芽苗的伸长生长有促进作用,继代次数影响芽苗的增殖分化,以继代6代以内最好。 4 生根培养:瓶内生根培养基1/2MS+IBA0.5mg/L+BA0.2mg/L+糖30g对双色茉莉的生根较好,生根率88.16%,平均根长4.96cm。瓶外生根为1/2蛭石+1/2珍珠岩+ASD5mg/L,生根率35.76%,平均根长6.49cm。 5 炼苗与移栽:有根双色茉莉移栽成活率高于无根苗,经强光闭瓶锻炼有利于提高移栽成活率,成活率达83.24%。炼苗适合的基质是蛭石:珍珠岩为1:1。 The stem of Brunfesia latifolia was used as explant to establish the technique systm of tissue culture.With Orthogonal design and analysis of variance,, we wanted to find out the best medium composition and the culture process in Brunfesia latifolia. The results were as follows:(1) As inoculation explant ,the middle stem of Brunfelsia latifolia in April ,May and Octomber was better than the other parts of the stem(2) Iniation culture: medium with l/2MS,IBA0.1mg/L ,NAA0.01mg/L ,BA3mg/L and KT2mg/L was the optimal.(3 ) Subculture: With MS,IBA0.5mg/L,BA3mg/L and actived carbon 0.2% as subculture medium, the increasing coefficient in 35d was 5.92.and nurslings above 2cm were 47.16% of the differentiated total buds. Actived carbon could prompt the growth of buds. Subculture generation had an influence on propagation and differentiation of buds. In general, the sixth generation was the best.(4) Rooting culture: For rooting culture, medium with MS,IBA0.5mg/L,BA3mg/L and sugar 30 g was the best with rooting rate 88.16% and average length of root 4.96cm, while rooting substrate with 1/2 perlite ,1/2 vermiculite and ASD5 mg/L was the best, rooting rate35.76% and average length of root6.49cm.(5) Transplantion: In transplanting, the survival rate of Brunfelsia latifolia roots was higher than that of Brunfelsia latifolia with no roots . Through strong illumination and in closed bottle, the survival rate could be improved to 83.24% .Substrate with perlite and vermiculite by 1 to 1 was the optimal.
